Abstract | Because of the recent Covid-19 Pandemic, people had used the antimicrobial agents extensively and in many times without prescription. Now it is expected that many antibacterial resistances will occur, for this reason this study focused on the discovering of new antimicrobial agent from natural sources, because it's very rich media in microorganisms that producing antimicrobial agents. The Dead Sea area in Jordan was chosen to study their soil bacteria, because it is a harsh environment due to its high temperature and the salinity of the sea area. In this study we successfully isolated three isolates with ability to produce antibiotic, these are coded (GH15, GH20, GH21). The activity of the three isolates was recorded, against gram positive and gram-negative bacteria. The supernatant of the isolates was extracted with hexane and ethyl acetate respectively. After the pilot study GH15, was chosen to complete the characterization studies because of the high potential to produce the desired antibiotic. The activity was detected in both hexane and ethyl acetate extracts. TLC and bioautography for both extracts were studied and the MIC and MBC of gram positive and gram-negative bacteria were determined. The results of this study are promising, the isolated antibiotic have broad spectrum and further testing is required to determine the full characteristic of the antibiotic and the producing bacteria. |
