Formulation optimization of polyamide disulfide disulfide nanoparticles of cromolyn by factorial design

The importance of using nanoparticles as drug carriers is due to their high carrier capacity, high stability, probability of incorporation of both lipophilic and lipophobic drugs, and probability of various routes of administration, including inhalation and oral routes. This study involved the preparation of cromolyn-polyamide-disulfide to nanoparticles overcome the problem of cromolyn frequent dose. The cromolyn-polyamide-disulfide samples were prepared using various amounts of both cromolyn (0.05, 0.10 and 0.20 g) and polymer (0.05, 0.10 and 0.20 mg) at different pH values (2.2 and 4.4) with different amounts of ferric chloride FeCl3 (0.6, 0.3 and 1.2 g). The produced nanoparticles were characterized by Fourier Transform Infrared (FTIR) and in vitro release. FTIR assessments were performed to estimate the functional group(s) of cromolyn-loaded polyamide-disulfide nanoparticles. Full factorial design (Minitab 18) was used for the purpose of analyzing the results by using graphical analysis such as surface and contour plots, Pareto chart, main effect plots, normal probability plot of the residuals and residuals versus corresponding predicted values plots and interaction plots. The independent variables were cromolyn, polymer, FeCl3 and pH, while the loading efficiency, zeta potential and particle size were the dependent variables. Through experiments, we were able to obtain nanoparticles to carry and preserve the drug and provide Prolonged release of cromolyn.
