Matrix metalloproteinase (MMPs), a zinc and calcium dependent protease that monitors the breakdown of the extracellular matrix (ECM), aids in maintaining homeostasis. Studies have shown that UV rays cause an imbalance in the turnover of the ECM, which leads to skin wrinkling and premature skin aging. UV rays also increase the expression of (MMPs), specifically MMP-1, in the skin which implies that developing highly effective MMPs inhibitors for topical application could have positive influences on photoaging and be a major goal in skin aging treatment. In this study, various chemical scaffolds were created with the goal of producing novel MMPs inhibitors. Molecular docking is used to investigate the inhibitory effects of a novel synthesized tocopherol derivatives on human fibroblast collagenase. For each of the MMPs proteins chosen, the designed compounds with the highest docking scores were compounds 3D, 3C, and 3A for MMP-1, MMP-8, and MMP-13 respectively. To achieve the highest yield, these derivatives were produced using different procedures. All the synthesized compounds showed good inhibitory activity against MMP1, 8, and 13. According to the data, 3D had a binding energy of 153.613, which was higher than the reference ligand's (129.805), 3C scored (168.226), and the reference ligand (154.287), while 3A exhibited (190.924) score although the reference ligand had a binding energy of (163.191). Every designed compound has a docking score that is higher than the docking score of the reference compound. Therefore, these novel compounds h ave a great potential to become skin-care products for preventing skin photoaging.
